The Neisser stain is a microbiological stain for visualising the polar bodies in the cytoplasm of gram-positive bacteria. This staining is used to identify diphtheria bacteria.
Cells are stained with methylene blue and crystal violet in acidic solution and then counterstained with chrysoidin solution. Polyphosphate granules appear black-violet in the yellow-brown stained cells.
